cells into the bloodstream, where circulating tumour cells (CTCs) survive for about 1 to 2.5 hours.<\/li>\n<li>In order to detach from the primary tumour and spread into the blood, the cells must undergo a cellular process known as epithelial-mesenchymal transition (EMT).<\/li>\n<li>EMT increases the migratory capacity of tumour cells, allowing cells to penetrate the vascular system and circulate as single or clusters of circulating tumour cells.<\/li>\n<\/ul>\n<ul>\n<li>CTCs exit the bloodstream after undergoing a reverse process called mesenchymal-epithelial transition (MET) and are able to colonise distant organs.<\/li>\n<\/ul>\n<ul>\n<li>CTCs are defined as EpCAM (+), PanCK (+), CD45 (-) cells. Circulating tumour-associated cells (C-TACs) are defined as EpCAM (+), PanCK (+), CD45 (+\/-) cells of tumour origin in the peripheral blood. <\/li>\n<\/ul>\n<ul>\n<li>Non-tumour cells in the peripheral blood have a functional apoptotic mechanism, but CTCs and C-TACs are resistant to apoptosis.<\/li>\n<\/ul>\n<ul>\n<li>An epigenetically active stabilisation process can eliminate normal cells and allow apoptosis-resistant CTCs and C-TACs to survive.<\/li>\n<\/ul>\n<ul>\n<li>Sufficient C-TACs can be enriched and obtained for immunocytochemical (ICC) profiling with markers used in immunohistochemistry (IHC), which helps in the histopathological subtyping of tumour tissue.<\/li>\n<\/ul>\n<ul>\n<li>To increase the sensitivity of the method, circulating free DNA (cfDNA) is tested at the same time. In very rare cases, no CTCs can be found, but cfDNA can be detected, leading to a positive result. However, cfDNA alone cannot provide any information as to the origin of the tumour disease.
